The primary focus of our lab is to understand the role of mitochondrial dynamics in cellular physiology, development and human disease. In addition, we study other aspects of mitochondrial biology, including organellar quality control and biogenesis.

Guan’s pioneering work with mitochondrial diseases included the discoveries of the mitochondrial cause of maternally inherited non-syndromic and aminoglycoside induced hearing loss. Currently, Dr. Guan's lab is focusing on investigating the mechanisms underlying the aberrant mitochondrial tRNA metabolisms including the synthesis, processing, maturation, CCA addition, posttranscriptional nucleotide modification and aminoacylation of tRNA, and their impact on human diseases including deafness, optic neuropathy and hypertension.

With a broad background in clinical neurology, cell biology, and biochemistry, he has contributed significantly to the field of mitochondrial disorders, neurodegeneration, and mitochondrial biochemistry and physiology. Dr. Manfredi has authored over 150 papers in peer-reviewed journals, including Hum Mol Genet, Nat Genet, J Biol Chem, J Cell Biol, Cell Metab, Brain, J Neurosci and Mol Biol Cell.

Human genetics is often employed to verify our research findings.
